چکیده

(Shaked etal., PLoS ONE 2008). To further investigate this finding, several different approaches have been taken. First, the expression of HDAC-regulated candidate genes responsible for controlling neurogenesis has been examined through quantitative realtime RT-PCR. TSA-based inhibition of HDAC activity resulted in upregulation of Bmp2, Stat1, Stat3, Ngn1 and Ngn2 in neural precursors from both cortex and striatum. In contrast, the expression of Smad7, an inhibitor of BMP2/4 signaling, decreased as a result of HDAC inhibition. This suggested that HDACs control neurogenesis by inhibiting the BMP2/4 signaling pathway, with additional involvement of JAK/STAT signaling and the Ngn transcription factors. Second, in order to ascertain which HDAC family members influenced neurogenesis, the expression of individual HDAC genes was inhibited via RNA interference. Expression of HDAC 1, 2, 4 and 5 could be effectively downregulated, as seen on the mRNA level. The effect of the knockdown on neurogenesis and astrogliogenesis will be elucidated. Third, the expression patterns of individual HDACs in developing brain tissue will be presented.

Genome-wide identification of Smad/Foxh1 targets reveals a role for Foxh1 in retinoic acid regulation and forebrain development.

Foxh1, a Smad DNA-binding partner, mediates TGFbeta-dependent gene expression during early development. Few Foxh1 targets are known. Here, we describe a genome-wide approach that we developed that couples systematic mapping of a functional Smad/Foxh1 enhancer (SFE) to Site Search, a program used to search annotated genomes for composite response elements. Ranking of SFEs that are positionally c...

Aberrant expression of homeobox gene SIX1 in Hodgkin lymphoma

In Hodgkin lymphoma (HL) we recently identified deregulated expression of homeobox genes MSX1 and OTX2 which are physiologically involved in development of the embryonal neural plate border region. Here, we examined in HL homeobox gene SIX1 an additional regulator of this embryonal region mediating differentiation of placodal precursors. SIX1 was aberrantly activated in 12 % of HL patient sampl...

A homozygous mutation in HESX1 is associated with evolving hypopituitarism due to impaired repressor-corepressor interaction.

The paired-like homeobox gene expressed in embryonic stem cells Hesx1/HESX1 encodes a developmental repressor and is expressed in early development in a region fated to form the forebrain, with subsequent localization to Rathke's pouch, the primordium of the anterior pituitary gland. Mutations within the gene have been associated with septo-optic dysplasia, a constellation of phenotypes includi...

HESX1- and TCF3-mediated repression of Wnt/β-catenin targets is required for normal development of the anterior forebrain.

The Wnt/β-catenin pathway plays an essential role during regionalisation of the vertebrate neural plate and its inhibition in the most anterior neural ectoderm is required for normal forebrain development. Hesx1 is a conserved vertebrate-specific transcription factor that is required for forebrain development in Xenopus, mice and humans. Mouse embryos deficient for Hesx1 exhibit a variable degr...

Transcription factor and microRNA interactions in lung cells: an inhibitory link between NK2 homeobox 1, miR-200c and the developmental and oncogenic factors Nfib and Myb

BACKGROUND The transcription factor NK2 homeobox 1 (Nkx2-1) plays essential roles in epithelial cell proliferation and differentiation in mouse and human lung development and tumorigenesis. A better understanding of genes and pathways downstream of Nkx2-1 will clarify the multiple roles of this critical lung factor. Nkx2-1 regulates directly or indirectly numerous protein-coding genes; however,...
